Kondisi ini mendorong pencarian alternatif terapi berbasis bahan alami, seperti madu dan kurma, yang diketahui memiliki kandungan senyawa bioaktif dengan potensi antibakteri. Penelitian ini bertujuan untuk membandingkan efektivitas antibakteri madu, ekstrak kurma, serta kombinasi keduanya terhadap pertumbuhan E. coli secara in vitro. Penelitian ini menggunakan desain eksperimental laboratorik dengan metode difusi cakram pada media Mueller Hinton Agar (MHA). Sampel terdiri dari kelompok perlakuan madu, ekstrak kurma, kombinasi keduanya, serta kontrol. Aktivitas antibakteri dinilai berdasarkan diameter zona hambat yang terbentuk, kemudian direncanakan untuk dianalisis secara statistik menggunakan uji non-parametrik Kruskal-Wallis dan dilanjutkan uji post hoc. Hasil penelitian menunjukkan bahwa seluruh kelompok perlakuan tidak memiliki aktivitas antibakteri terhadap E. coli, yang ditandai dengan tidak terbentuknya zona hambat pada seluruh perlakuan. Oleh karena itu, data yang diperoleh tidak memenuhi syarat untuk dilakukan analisis statistik sehingga tidak dilakukan uji statistik lebih lanjut. Kesimpulan dari penelitian ini adalah madu, ekstrak kurma, dan kombinasi keduanya tidak menunjukkan efek antibakteri terhadap E. coli dalam kondisi penelitian ini.
