Penelitian ini menggunakan metode penelitian eksperimental, yang kemudian terbagi menjadi kelompok kontrol serta kelompok perlakuan dengan variasi konsentrasi ekstrak 39,06; 78,125; 156,25; dan 312,5 μg/mL. Karakterisasi senyawa aktif fitokimia dianalisis menggunakan instrumen Gas Chromatography-Mass Spectrometry (GC-MS) yang memperlihatkan dominasi senyawa golongan terpenoid dan ester asam lemak. Efek perlakuan dievaluasi berdasarkan pengamatan kualitatif perubahan morfologi sel menggunakan mikroskop inversi yang memperlihatkan adanya tanda kerusakan sel akibat paparan ekstrak berupa gejala pengerutan (cell shrinkage), perubahan bentuk menjadi membulat (cell rounding), serta akumulasi debris sel seiring peningkatan konsentrasi perlakuan. Uji viabilitas kuantitatif dilakukan dengan metode MTT assay dan di lakukan analisis data menggunakan uji One-Way ANOVA dan analisis regresi linear untuk menentukan nilai Inhibitory Concentration 50 (IC50). Hasil penelitian menunjukkan bahwa ekstrak metanol daun Rasamala mampu menurunkan viabilitas sel MCF-7 secara signifikan seiring peningkatan konsentrasi perlakuan (dose-dependent manner) dengan p < 0,0001. Nilai IC50 ekstrak metanol daun Rasamala terhadap sel MCF-7 diperoleh sebesar 191,67 μg/mL yang menunjukkan aktivitas sitotoksik kategori sedang (moderate cytotoxic activity).
