TERHADAP ERα SEBAGAI ANTI KANKER PAYUDARA Rahman Sadli Waraiya 1504015310 Kanker payudara merupakan penyakit kanker yang paling sering diderita oleh wanita dan merupakan penyakit kanker dengan jumlah kasus baru terbanyak di dunia pada tahun 2020. Ekstrak Rosemari diketahui memilki potensi sebagai antikanker payudara karena menghambat ERα pada sel T-47D dan sel MCF-7 dengan IC50 masing-masing sebesar 38,3 ± 2,8 μg/mL dan 65,5 ± 8,7 μg /mL. Penelitian ini bertujuan untuk menguji afinitas dan kestabilan senyawa Rosemari terhadap ERα sebagai antikanker payudara dengan metode penambatan molekuler dan dinamika molekuler. Penambatan molekuler dilakukan menggunakan Autodock 4.0 dan dinamika molekuler menggunakan Gromacs. Hasil penambatan molekuler menunjukan Kariofilen Oksida memiliki nilai afinitas sebesar -7,70 kkal/mol sedangkan tamoxifen sebagai kontrol positif sebesar -10,33 kkal/mol. Hasil dinamika molekuler menunjukan kompleks protein Kariofolen oksida memiliki nilai energi potensial dan Root Mean Square Distance (RMSD) yang stabil sedangkan nilai Root Mean Square Fluctuation (RMSF), Radius of Gyration, ikatan hidrogen dan energi energi bebas (ΔG) dengan metode Molecular Mechanics Poisson–Boltzmann Surface Area (MMPBSA) menunjukan kompleks protein Kariofilen Oksida tidak lebih stabil dibandingkan kompleks protein tamoxifen.
