Kombinasi surfaktan akan menghasilkan niosom yang lebih baik dibandingkan pemakaian surfaktan lipofilik tunggal. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh nilai HLB kombinasi surfaktan Tween 60 dan Span 60 terhadap stabilitas fisik niosom Glukosamin HCl. Niosom dibuat menggunakan metode hidrasi lapis tipis dengan variasi nilai HLB 4,7 (F1), 6,15 (F2), 6,74 (F3), dan 8,1 (F4). Pengujian stabilitas dilakukan pada suhu 4oC dan 40oC selama 8 minggu yang meliputi uji organoleptis, pH, bobot jenis, zeta potensial, ukuran partikel, % PDI. Pada sistem niosom juga dilakukan pengujian morfologi dan efisiensi penjerapan. Niosom yang dihasilkan merupakan sistem dengan partikel berbentuk bulat dengan nilai efisiensi penjerapan 57,215 % (F1), 62,133 % (F2), 65,850 % (F3), dan 69,571 % (F4). Pada penyimpan suhu 4oC, niosom yang dihasilkan berbentuk suspensi berwarna putih yang homogen, sedangkan pada penyimpanan suhu 40oC, niosom mengalami perubahan warna menjadi coklat. Rata-rata ukuran partikel niosom adalah 271,83 – 631,85 nm dengan zeta potensial (-)36,67 – (-)63,94 mV, PDI 0,000,00 - 0,510,06 dan bobot jenis 1,006 – 1,045 g/mL. Variasi HLB kombinasi tween 60 dan span 60 tidak mempengaruhi stabilitas fisik niosom pada penyimpanan suhu 4 oC dan 40oC.
