SEBAGAI PENGHASIL ENZIM XILANASE DAN AMILASE ispublished: pub subjects: R subjects: RS divisions: 48201 abstract: Bakteri endofit hidup di dalam jaringan tanaman tanpa merugikan tanaman inangnya, dapat ditemukan pada semua jenis tumbuhan seperti pohon berkayu, salah satunya pohon asam jawa (Tamarindus indica L.). Penelitian ini bertujuan untuk skrining bakteri endofit ranting asam jawa yang memiliki aktivitas enzim xilanase dan amilase yang paling potensial. Isolasi bakteri endofit ranting asam jawa dilakukan dengan teknik penanaman langsung pada medium Nutrient Agar (NA). Uji aktivitas enzim secara kualitatif dilakukan menggunakan medium starch dan xilan dengan mengamati zona bening yang terbentuk di sekitar koloni bakteri endofit yang ditumbuhkan pada medium, kemudian uji aktivitas enzim kuantitatif dilakukan menggunakan metode Dinitrosalisilat (DNS) dengan spektrofotometer UV-Vis λ 535 nm. Isolasi bakteri endofit dari ranting asam jawa diperoleh 3 isolat yang dua diantaranya (AJ 2, AJ 3) menunjukkan kemampuan untuk menghasilkan enzim amilase dan isolat AJ 3 memiliki aktivitas enzim amilase terbesar yaitu 6,3317 U/ml. Namun ketiga isolat tersebut tidak dapat menghasilkan enzim xilanase.
