Secara khusus, mutu hasil belajar sains dipengaruhi oleh kegiatan praktikum yang dilakukan siswa di sekolah. Namun, kegiatan praktikum yang sudah berjalan selama ini belum banyak diselidiki. Studi ini di desain untuk mengeksplorasi persepsi siswa terhadap proses pembelajaran praktikum di Sekolah Menengah Atas (SMA) Negeri di Indonesia. Studi kuantitatif ini melibatkan 250 siswa dari 12 SMA Negeri untuk mengisi kuesioner secara sukarela. Penentuan partisipan dilakukan secara convenience sampling dengan kriteria telah melaksanakan kegiatan praktikum minimal 3 kali. 31 item pernyataan kuesioner skala Likert disusun berdasarkan empat indikator yaitu kesesuaian praktikum dengan materi, pelaksanaan kegiatan praktikum, penggunaan teknologi dalam kegiatan praktikum dan evaluasi kegiatan praktikum. Tanggapan responden ditabulasikan pada lembar Excel dan analisis lanjutan dengan Rasch Model melalui aplikasi Winstep 4.4.4. Temuan ini mengungkapkan pelaksanaan kegiatan praktikum biologi pada SMA Negeri sudah berjalan cukup baik. Siswa memiliki sikap tanggungjawab saat melaksanakan praktikum mulai dari penggunaan alat dan melakukan langkah kerja sesuai prosedur, serta mematuhi tata tertib laboratorium. Berdasarkan data demografi, siswa dengan jenis kelamin perempuan lebih memiliki persepsi yang baik terhadap disiplin waktu saat pelaksanaan kegiatan praktikum dibandingkan dengan siswa laki-laki.
