Daun lempuyang gajah (Zingiber zerumbet (L.) Roscoe ex Sm.) memiliki aktivitas penghambatan biofilm Streptococcus mutans. Penelitian ini bertujuan untuk mengetahui penghambatan biofilm Streptococcus mutans tertinggi dari hasil kromatografi kolom fraksi metanol daun lempuyang gajah. Penelitian ini diawali dengan maserasi bertingkat menggunakan pelarut n-Heksan (non polar), etil asetat (semi polar), dan metanol (polar), kemudian fraksi metanol yang diperoleh dielusi menggunakan kromatografi kolom dengan metode step gradient polarity (SGP). Hasil dari kromatografi kolom, selanjutnya dilakukan pemisahan berdasarkan Rf dengan metode Kromatografi Lapis Tipis (KLT). Hasil kromatografi kolom dengan Rf yang mendekati, kemudian dikelompokkan dan dilakukan uji penghambatan biofilm Streptococcus mutans. Hasil yang didapat menunjukkan bahwa hasil kromatografi kolom fraksi metanol daun lempuyang gajah pada rentang Rf 0,52-0,67 memiliki penghambatan biofilm Streptococcus mutans tertinggi dengan nilai IC50 sebesar 171,48 μg/ml dan potensi relatif 8,04 kali klorheksidin glukonat.
